An evidence based approach

Promoting social and emotional wellbeing is seen increasingly by policy makers as an important component of broader strategies designed to improve a broad range of health and related outcomes for children and young people. This focus has the potential to minimise the health risks and negative social outcomes associated with ‘modern’ childhood and adolescence, including anti-social behaviour, binge drinking, and lack of involvement in training and employment.

The life course approach provides a strong theoretical rationale for investing in promoting social and emotional wellbeing as evidence indicates that cumulative exposure to adverse social circumstances increases a child’s risk of emotional and behavioural problems during childhood, but also their health and social prospects in later life. Conversely, and most recently asset based approaches argue that the more opportunities we provide children and young people to accumulate a range of positive experiences the more likely they are ‘do well’ even in the face of difficult circumstances.

Over the last few years, the National Institute of Health and Clinical Excellence (NICE) has developed evidence based guidance to promote social and emotional wellbeing at 3 key life stages; early years (at home and early education), entering primary school and transitions to secondary education. The guidance defines the key features of effective programmes and the conditions necessary for their successful implementation.

 This paper describes the processes and methods used by NICE to produce evidence based guidance and the challenges faced in developing and accruing evidence that supports positive approaches to promote social and emotional wellbeing during early years, through adolescence and beyond.

The majority of new biopharmaceuticals are made in Chinese hamster ovary cells, a transformed cell type originally isolated in the 1950’s.  The adaptability and utility of CHO cell factories derives from our exploitation of their acquired genetic/functional variation using high-throughput functional screening and selection processes which enable industry to isolate and maintain cell factories with unusual and desirable properties.  However, we remain blind with respect to enabling cellular mechanisms, learning retrospectively not how to design, build and control intrinsically better cell factories, but only how to iteratively improve screening and process environments.  Can we apply the new concepts and tools of systems and synthetic biology disrupt this industrial “Groundhog Day” modus operandi, and enable application of the engineering design paradigm to the heart of the biomanufacturing process - the cell itself?

Health Behaviour in School-Aged Children (HBSC) is a cross-national study of the health of children, coordinated by the University of St. Andrews. Members of the Canadian HBSC team are visiting St. Andrews to discuss some new ideas that will inform their national as well as the international HBSC efforts. In this seminar, participants will be exposed to the theory of holistic child health, which suggests that composite in addition to more specific measures of health are important to understanding the health of children. Following the seminar, an open workshop will be held to discuss practical approaches to the measurement of holistic health in children. Ideas from this workshop will support ongoing HBSC research in Canada and other countries.

Synthetic biology is being used to build devices through both top-down and bottom-up approaches.1 For example, genome engineering has been used to reprogram cells, and DNA origami has been used to produce a variety of nanodevices. While progress has been made on the assembly of minimal cells, synthetic tissues have so far received limited attention.1 We have assembled networks of aqueous droplets joined by lipid bilayers.2 The droplets in the networks can communicate with each other and with the environment through engineered protein pores3 and, like tissues, exhibit emergent properties.4 To mimic tissues, droplet networks should be endowed with various properties including the ability to store and use energy, to move and change shape, to detect signals, to carry out computations and take up and release molecules. At a certain level, these goals have been achieved.2-5 We now aim to interface droplet networks with living tissues and control them with electrical or optical signals.

The human genome encodes over 500 protein kinases, which regulate specific process throughout the lifetime of the cell.  Protein kinases, therefore, are potential targets for new medicines.  I shall describe how knowledge of protein kinase structure-function relationships helps in the identification, evaluation and optimisation of inhibitors for use as drugs.  Several kinase inhibitors with different molecular mechanisms have been approved to treat cancer and I shall discuss how clinical data has improved our insight into the challenges and opportunities for kinase drug discovery.  I shall illustrate how identification of diverse kinase inhibitors, which have different chemical and biological properties, increases the chances of succes in finding new medicines.

As part of the 600 anniversary celebrations for the University of St Andrews we cordially invite you to a lecture given by Sir John Meurig Thomas, Honorary Professor at the Department of Materials Science and Metallurgy, Cambridge.

Sir John occupied the Chair of Chemistry created for Michael Faraday at the Royal Institution of Great Britain, of which he was Director (1986-1991). Formerly he was Head of the Department of Physical Chemistry, University of Cambridge (1978-1996) and Master of Peterhouse (1993-2002).

Lord Rutherford said of Michael Faraday that he was one of the greatest experimenters ever; Albert Einstein believed that Faraday was responsible for the greatest change in the intellectual structure of physics since Newton. There is little doubt that Faraday bequeathed a greater corpus of useful knowledge than any other physical scientist. How did it come about that Faraday, a deeply religious man who left school at thirteen to become an errand boy and then an apprentice to a bookbinder (a young man who never attended high school or a university and knew no mathematics), could reach such pinnacles? In terms that are intelligible to non-scientists and interested lay persons, the speaker shall endeavour to answer the above and related questions. The tale of Faraday’s achievements and discoveries is one of the most romantic in the history of science, his character, intellect and commitment speak to us through all ages.

The Wonder of the Wanderer (what the vagus nerve can do for you..)  

Established in the memory of Professor Sir George Lindor Brown (1903-1971), this is an annual series of lectures aimed to stimulate an interest in Physiology. The subject of this year's lecture is the vagus nerve, the 10th cranial nerve. This parasympathetic nerve originates in the brainstem, transmitting signals to and from many major organs in thebody, helping them perform their normal functions.   Prof. Deuchars will explore some of the functions of the vagus, concentrating on its involvement in disease states.Electrical stimulation of the vagus nerve is currently being investigated as a treatment for a surprising diversity of diseases. The evidence for this will be examined. This will be an interactive lecture with some audience participation, but no need to be nervous as not one individual will be singled out!

Is Earth the only planet with photosynthesis?

Abstract:

Astrobiology seeks (my definition) to set criteria for the occurrence of life other than on Earth, and to devise and employ methods for detecting any such life. I have chosen photosynthetically supported life because I know something about it, which may seem like looking for keys lost in a street at night under a street-lamp rather than where you think you lost them. However, in this case the street-lamp may be the right place, for two reasons. One is likeliehood: a diverse and productive biosphere must be energized by electromagnetic radiation; geochemical energy inputs to Earth-like planets are inadequate. The other is detectability: use of the commonest reductant in the habitat of life as we know it, i.e. water, generates oxygen in larger quantities than can be generated geochemically (Lovelock's test), and oxygen (as oxygen or ozone) and the equivalent of the reflectance "red edge" of chlorophyll on Earth can potentially be (very) remotely sensed by reflectance spectroscopy. In the lecture I shall attempt to justify these assertions, and also discuss recent work with colleagues on the potential for photosynthesis in (astronomically) reflected radiation, on planets in binary star systems, and for "cryptic photosynthesis" where products, but not pigments, could be remotely sensed.

Professor Moussa Youdim

 

Professor Youdim holds the Finkelstein Chair in Life Sciences and Pharmacology at the Technion Israel Institute of Technology in Haifa, and is the director of the Eve Topf and U.S. National Parkinson Foundation Center of Excellence for Neurodegenerative Diseases Research and Teaching. He is also Distinguished Professor at Hong Kong and Hong Kong Poly Universities. He is known internationally for his research and drug development activities in depressive illness and Parkinson's and Alzheimer's diseases.

 

Professor Youdim is a world expert in neurodegenerative disease research. He is named on more than 110 international patents and has been associated with the development of successful drugs during his career such as rasagiline (Azilect®), approved for treatment of Parkinson's disease in 27 European countries, Canada and the United States, and ladostigil, a candidate drug in Phase II clinical trials for Alzheimer's disease. He has published more than 500 peer-reviewed articles, 45 books and more than 300 review articles. He serves on the editorial boards of many journals of neurochemistry, pharmacology, neuropharmacology and neuropsychiatry, and has received more than 50 national and international awards.

 

From chemistry through crystal structures and in vivo effects to drug development for neurodegenerative diseases, this seminar will have something for everyone!

Abstract: The S-adenosyl methione (SAM) riboswitch is one of the most frequently occurring riboswitch and has been shown to control transcription and translation.  The aptamer domain is organized around a four-way junction and the global structure is characterized by the formation of two sets of coaxially helical stacks, P1-P4 and P2-P3.  Using CGE and FRET, we characterized the metal ion- and SAM-induced folding of the SAM riboswitch and found that the aptamer undergoes an ion-induced folding as well as a SAM-induced transition, which is characterized by a large P4 stem structural change.  Our findings provide a first rationale about how the riboswitch architecture is folded as observed in the crystal structure.

In the aptamer, the P4 stem is the only peripheral element for which no obvious role can be deduced from previous studies.  To understand how the P4 stem influence the SAM riboswitch activity, we characterized a variant found in Fusobacterium, which does not bear a P4 stem.  By using 2-aminopurine fluorescence, CGE, FRET and single-round in vitro transcription assays, we find that the P4 stem acts as an ligand affinity modulator, which can fine-tune the SAM affinity only by a matter of adding or removing base pairs.  This suggests that riboswitches may rely on peripheral elements as well as core variations to alter ligand affinity, which is crucial to allow specific gene regulation.  We are currently characterizing the influence of P4 on the folding dynamics using single-molecule FRET.

Ken Murray will be receiving an honorary degree at the Science Graduation this week.  His first interests were in nucleic acids and nucleoproteins and then in bacteriophages and along with his wife Noreen developed restriction enzyme technology thus playing a major role in the initiation of genetic engineering.  Ken used this experience to construct a very effective Hepatitis B vaccine which is still in worldwide use.  He was one of the original founders of Biogen.

Professor Sue Black is head of the Centre for Anatomy and Human Identification at the University of Dundee. She graduated from Aberdeen University and after a spell working at St.Thomas' and Guys Medical Schools in London she returned to Scotland and joined Dundee in 2003. She was awarded an OBE for her services to forensic anthropology in Kosovo following the Balkan crisis and she has worked in Iraq, Sierra leone and Thailand. She is the founder of the Centre for International Forensic Assistance and the British Association for Human Identification. She is specialty assessor in forensic anthropology for the Council for the Registration of Forensic Practitioners and is a Fellow of the Royal Society of Edinburgh.
